A quality by design approach for optimization of Lecithin/Span® 80 based nanoemulsions loaded with hydrophobic drugs

Abstract Lately, nanoemulsions loaded with hydrophobic drugs have been successfully developed to improve the treatment of several global diseases. On this subject, a detailed study crucial role excipients and experimental conditions used for these nanosystems is still required. Thus, aim work was development Benzidazole (Class I, log P = 0.91), Praziquantel II, P = 2.44), Pyrimethamine II/IV, P = 2.69), Niclosamide P = 4.5), Triclabendazole P = 5.9) using Span® 80, soybean lecithin Miglyol® 812 as excipients. A Placket-Burman design selected identify main parameters that influence in desirable characteristics such formulations. Then, full factorial built analyze effect factors identified screening phase. Plackett-Burman indicated were two most influencing on hydrodynamic diameter systems. In addition, association efficiency influenced by P each drug while response stability PBS modified 80 P. The results revealed concentration values remarkable impact nanosystems. optimal preparation verified other independent experiment agreement predicted optimum values. methodology could serve an attracttive platform deliver hydrophobics compounds stable nanoemulsions.
